Rome alone deserves three days minimum if you want to do it properly, not just check a box. Florence is two days at least. Venice, one to two. Amalfi and Cinque Terre each require a day of travel to reach, and the Amalfi Coast roads in October can be slow with buses and tour groups.
What you have described would leave everyone exhausted, and your in-laws would likely struggle with the pace.
October in Tuscany is one of the best months in Italy. Harvest season, no summer crowds, the light is extraordinary. Your teens will actually enjoy it. Your in-laws can pace themselves. We can find a private villa for two or three nights that gives everyone their own space.
Cinque Terre in October is still beautiful and far less crowded than summer. Amalfi is still accessible. I would pick one based on what your family cares about more, dramatic coastal scenery or hiking and village life.
